Antibiotic resistance

XL1B cells are resistant to tetracycline

Transformation protocol

 

1. Thaw the cells on ice

2. Add experimental DNA to the cells and gently mix

3. Incubate the tube on ice for 1~10 minutes

4. Heat-pulse the tube for 45 seconds at 42℃

5. Incubate the tube on ice for 2 minutes

6. Add 500ul LB medium and incubate for 30 minutes at 37℃

7. Plate cells on LB agar plate containing the appropriate antibiotic

8. Incubate the plate at 37℃ overnight
